Fenbendazole is a benzimidazole anthelmintic used to treat helminthic infections in different species. It has been approved by the U.S. Food and Drug Administration for use in food-producing animals in 1978 and is now widely used against praziquantel-resistant schistosomiasis in humans through its addition to the drinking water of infected individuals. It is available as tablets, feed additive, and injectable formulations for domestic animals, which are approved for human use.
Fenbendazole capsules has been shown to selectively bind to glutamate-gated chloride channels, inhibiting the synaptic release of gamma-aminobutyric acid (GABA) and inducing a calcium influx into target cells. This effect leads to disruption of neuronal signaling and paralysis of parasites. It was found also that fenbendazole blocks voltage-dependent calcium channels, preventing transmission at the neuromuscular junction when tested in frogs.
fenbendazole capsules is not approved for human use in the United States, but is commonly used off-label to treat neurocysticercosis.
Benzimidazoles can be toxic in humans, similarly to their mechanism of action (disruption of the GABA neurotransmitter system), benzimidazoles tend to inhibit acetylcholinesterase activity. At therapeutic doses, about half of patients develop at least mild sedation/drowsiness, with some also experiencing muscle weakness and slurred speech or blurred vision. At higher doses (more than 10 times the therapeutically effective dose), more severe symptoms may occur including convulsions and coma. |
